The physics of lyophilization couples heat transfer, mass transfer, and phase behavior. Sublimation requires a vapor pressure difference between the ice front and the chamber, and the dried layer adds resistance to vapor flow. Amorphous formulations are characterized by a glass transition temperature of the maximally freeze-concentrated solute, often denoted Tg'. Crystalline bulking agents can provide structure, while amorphous excipients stabilize labile components. Open questions remain about spatial heterogeneity, edge effects, and how laboratory cycles scale to production.
Lyophilization is a drying process in which a solvent, usually water, is removed from a frozen material by sublimation under reduced pressure. The material is first solidified, then placed under vacuum so that ice transitions directly to vapor without a bulk liquid phase. This approach suits heat-sensitive substances that would degrade during conventional evaporation. Primary drying removes unbound ice, while secondary drying reduces water that remains adsorbed to the solid matrix. The result is a porous, lightweight solid that can be reconstituted later.
A typical cycle begins with freezing, sometimes including an annealing step to control ice crystal size. Freezing conditions influence the pore network that later allows vapor escape. During primary drying, shelf temperature and chamber pressure are set so heat enters the product while its temperature stays below the collapse or eutectic point. Secondary drying then raises the shelf temperature to desorb bound water and lower residual moisture. Cycle design depends on formulation, fill volume, container type, and equipment capability.
Misconceptions about lyophilization include the idea that dried products are indefinitely stable or that vacuum sealing eliminates all degradation. Chemical reactions can continue in the solid state, and some proteins lose activity even at low moisture. Another misconception is that any freeze-dryer cycle can be scaled by time alone; heat and mass transfer differ with equipment and load. Open questions remain about predicting long-term stability from short accelerated studies, particularly for amorphous formulations. Real-time stability data remain the standard for shelf-life assignment.
After lyophilization, the dried product is often sealed under vacuum or an inert gas to limit moisture and oxygen exposure. Vials, stoppers, and seals must maintain their barrier throughout shelf life. Storage temperature depends on product sensitivity: some cakes tolerate controlled room temperature, while labile biologics require refrigeration. Humidity is a critical variable because dried cakes are hygroscopic and can absorb water when exposed to air. Handling procedures therefore limit open-vial time and use desiccated environments for sampling.

The physics of lyophilization depends on phase boundaries and heat and mass transfer. During primary drying, heat supplied to the product must equal the latent heat of sublimation, while water vapor moves through the drying cake to the condenser. If shelf temperature or pressure is set too high, the ice front can exceed the collapse temperature, causing meltback or pore collapse. If conditions are too cold, drying slows and costs rise. Formulation excipients, vial geometry, and freezing rate alter these limits.
Equipment for lyophilization includes a vacuum chamber, temperature-controlled shelves, a condenser, and a vacuum pump. A refrigeration system cools the shelves and condenser below the product's freezing point. Process monitoring often uses Pirani and capacitance manometers, thermocouples, and resistance sensors. Cycle development balances product quality with time and energy use. Some products are annealed during freezing to improve crystallization of bulking agents. Open questions remain about scaling cycles between laboratory, pilot, and production freeze-dryers.
Lyophilization, also called freeze-drying, removes water from a frozen material by sublimation under reduced pressure. The process begins with freezing, which converts free water into ice and may also produce a glassy phase. Primary drying then lowers chamber pressure so ice sublimes directly to vapor without passing through a liquid stage. Secondary drying raises the temperature modestly to remove bound water. The result is a porous, dry solid that usually reconstitutes quickly. Each stage influences pore structure, residual moisture, and stability.
After lyophilization, a product's quality depends on residual moisture, cake appearance, and reconstitution time. Residual moisture is often measured by Karl Fischer titration or thermogravimetric analysis. A low moisture content can slow chemical degradation, but overly dry cakes may be brittle or slow to dissolve. Stability studies track these attributes over months under defined temperature and humidity conditions. Batch records link these measurements to specific process runs and help identify trends before a product fails specification.
Storage conditions for dried products usually aim to exclude moisture and oxygen. Vials are sealed under vacuum or with an inert gas, and stoppers must maintain a barrier during transport. Temperature recommendations vary; some materials remain stable at room temperature, while others need refrigeration or frozen storage. Humidity control is critical because dried cakes can absorb water rapidly once a container is opened. Desiccant packs and moisture-barrier bags add further protection during shipping.
Such systems may use taxonomic thesauri to track accepted names and synonyms and to keep checklists aligned with changing taxonomy. Hardisty and colleagues have proposed the concept of "digital extended specimens" as an infrastructure for linking biodiversity records derived from physical specimens to a broader network of related digital objects and third-party data resources. Digitization improves access to specimen data, but it cannot by itself resolve cryptic diversity, which often requires direct examination and sequencing of preserved material. Despite these advances, many regions, particularly in the tropics, still lag behind in collecting and digitizing fungal diversity.
The freezing point of a solution is lower than that of the pure solvent, and the freezing-point depression (ΔT) is directly proportional to the amount concentration for dilute solutions. When the composition is expressed as a molality, the proportionality constant is known as the cryoscopic constant (Kf) and is characteristic for each solvent. If w represents the mass fraction of the solute in solution, and assuming no dissociation of the solute, the molar mass is given by
=== Cordelia Hanani-Spyrka === Cordelia Hanani-Spyrka (Claire Forlani) is Yasmin's aunt and the sister of her late father Charles, working as a public relations executive. Yasmin invites her to Henry's 40th birthday, where Cordelia gushes about her relationship with a younger man, and advises Yasmin not to tie her future to Henry, warning that men invariably weaponize women's unconditional love. Yasmin later catches Cordelia performing oral sex on Otto Mostyn and confronts her afterwards, asking why she never came to Charles' funeral and whether she knew about his serial sexual predation; Cordelia dismisses her brother's behavior, arguing that both she and Charles came from a "bohemian childhood", and it is implied the two had an incestuous relationship in their youth. Before leaving, Cordelia spitefully tells Yasmin that Charles planned to terminate her before learning she would be born a girl.
In 2000, Guinness's 1999 advertisement Surfer was named the best television commercial of all time, in a UK poll conducted by The Sunday Times and Channel 4. This advertisement is inspired by the famous 1980s Guinness TV and cinema ad, Big Wave, centred on a surfer riding a wave while a bikini-clad sunbather takes photographs. The 1980s advertisement not only remained a popular iconic image in its own right; it also entered the Irish cultural memory through inspiring a well-known line in Christy Moore's song "Delirium Tremens" (1985). Surfer was produced by the advertising agency Abbott Mead Vickers BBDO; the advertisement can be downloaded from their website. Guinness won the 2001 Clio Award as the Advertiser of the Year, citing the work of five separate ad agencies around the world. In 2002, Guinness applied the Michael Power formula to Asia with the character Adam King. The campaign featured such tag lines as: "Everyday someone, somewhere achieves something new. Sometimes on a grand, dramatic scale. Sometimes on a more personal scale." As of 2004, Guinness ranked among the top three beer labels in Singapore and Malaysia, with a 20 per cent market share across Southeast Asia. Malaysia was the brand's third-largest market in the region and the sixth largest market worldwide. In 2003, the Guinness TV campaign featuring Tom Crean won the gold Shark Award at the International Advertising Festival of Ireland, while in 2005 their Irish Christmas campaign won a silver Shark.
Episode Two opens with Alyx and Gordon learning a superportal to the Combine dimension has formed in the Citadel's place, progressing to a stage where the Combine can send reinforcements. They also discover that Alyx's encrypted data from the Citadel can reverse the portal, and so traverse the countryside to deliver the packet to another resistance headquarters at White Forest. As they progress, Combine Advisors have escaped the Citadel's destruction, and remaining Combine forces are regrouping, albeit under attacks by Vortigaunts. Aware of the resistance's plans to close the superportal, the Combine attack White Forest in force, but are repelled. The Combine are again the primary antagonist in the prequel Half-Life: Alyx, taking place between Half-Life and Half-Life 2. The game focuses on the efforts of Alyx Vance, her father Eli, and fellow resistance member Russell, as they attempt to infiltrate a massive Combine vault, believing it possesses a weapon that they could use to weaken the Combine occupation on Earth. After navigating through various quarantine zones of City 17 and rescuing Eli, they discover that the Vault is constructed not to hold a weapon, but as a prison, which they deduce to be holding Gordon Freeman. Alyx infiltrates the Vault and learns that it does not imprison Freeman, but is instead harboring the mysterious G-Man, who shows Alyx a glimpse of Eli's death in the future and offers her a chance to prevent it, which she accepts, before placing her in stasis.

=== Calcium-sensing receptor (CaR) === The secretion of parathyroid hormone (PTH) is regulated by the interaction of the calcium-sensing receptor with calcium in the blood. The calcium-sensing receptor is present on the plasma membrane of the chief cells. The CaR is a G protein-coupled receptor, as part of the C family. The CaR is divided into three general domains. These include an NH2-terminal extracellular end, a COOH-terminal intracellular end, and seven transmembrane domains. The CaR interacts positively with phospholipase C (PLC) and adenylyl cyclase. The CaR includes phosphorylation sites for protein kinase C (PKC) and protein kinase A (PKA). The phosphorylation of the PLC is seen to inhibit the secretion of PTH due to high calcium levels in the blood. The function of the PKA sites is currently unknown.
=== Territory === Bophuthatswana had a surface area of approximately 40,000 km2 (15,000 sq mi) and consisted of seven enclaves dispersed over the former South African provinces of Cape Province, Transvaal, and Orange Free State. Six of the enclaves were located relatively close together with three in the Cape Province and another three in Transvaal. The seventh enclave was in the Orange Free State between Bloemfontein and the Lesotho border. One of the enclaves shared a border with Botswana, while two of the enclaves were located near Pretoria and its surrounding industrial areas. The townships in these enclaves, such as Ga-Rankuwa and Mabopane continued to serve as dormitory townships for the supply of labour (as they had done prior to Bophuthatswana's independence) despite being located in Bophuthatswana territory. Other enclaves were similarly located near South African cities such as Rustenburg and Bloemfontein. The capital, Mmabatho, was situated in the enclave bordering Botswana. The territory and borders of the country were fluid as the South African government frequently incorporated territory into the country. As a result, when independence was declared in 1977, the country originally consisted of six enclaves but just before its reincorporation into South Africa, it had seven enclaves. Another example was the incorporation of Mafeking, which was located just outside the borders of Bophuthatswana when it gained independence in 1977, into the country in 1980 after a local referendum.
The larvae will push out the forming juvenile sand dollar and begin metamorphosis. For sand dollars, this stage takes about an hour and thirty minutes for the larval tissues to regress. Since the larval tissues are in regress, the sand dollar will then start to undergo resorption of these tissues. After metamorphosis, the sand dollar will start to become a juvenile, developing a skeleton, teeth, and will begin to be able to feed themselves.
== History == Sarcosine was first isolated and named by the German chemist Justus von Liebig in 1847. Jacob Volhard first synthesized it in 1862 while working in the lab of Hermann Kolbe. Prior to the synthesis of sarcosine, it had long been known to be a hydrolysis product of creatine, a compound found in meat extract. Under this assumption, by preparing the compound with methylamine and monochloroacetic acid, Volhard proved that sarcosine was N-methylglycine.

Over the years, multiple synthesizers have been developed to assist with automated synthesis, including the Chemspeed Accelerator (SLT106, SLT II, ASW2000, SwingSLT, Autoplant A100, and SLT100), the Symyx system, and Freeslate ScPPR. Recently, researchers have investigated the optimization of these methods for controlled/living radical polymerization (CLRP), which faces issues with oxygen intolerance. This research has led to the development of oxygen-tolerant CLRP, including with the use of enzyme degassing of RAFT (Enz-RAFT), atom-transfer radical (ATRP) that possesses tolerance to air, and photoinduced electron/energy transfer–RAFT (PET–RAFT) polymerization. Through the use of liquid-handling robots, Tamasi et al. demonstrated the use of automated synthesis with executing multi-step procedures, enabling the reactions to investigate more elaborate schemes, such as with scale and complexity. Lee Cronin and his team have developed a modular synthesis machine called the chemputer which uses a dedicated programming language for chemical synthesis.

Freezing only converts liquid to solid. Lyophilization adds vacuum and controlled warming so frozen solvent sublimes, leaving a dry porous solid. The two steps are related but not interchangeable.
Reduced pressure keeps the solvent below its triple point, allowing ice to become vapor without melting. Vacuum also helps remove water vapor from the product chamber. The exact pressure is chosen with the formulation and equipment.
Residual moisture is water that remains in the dried solid after secondary drying. It is often measured by Karl Fischer titration, near-infrared spectroscopy, or thermogravimetry. Acceptable levels depend on the material and its stability profile.
No. Freeze-drying removes water but does not reliably kill microorganisms. Sterile lyophilized products are typically prepared aseptically before freezing or are sterilized by a validated method. Microbial control depends on the entire manufacturing process.
